Note the Panel B-middle image, which shows intense nuclear clustering of ionizing radiation-induced phosphorylation of ATM at serine 1981. In contrast, Panel A-middle image shows no detectable phosphorylated ATM.

Background

ATMAtaxia Telangiectasia Mutated is a PI3-family protein kinase and a critical cell cycle checkpoint protein. ATM and a related protein, ATR, play crucial roles in the maintenance of genome integrity and the response to DNA damage. Upon activation, ATM phosphorylates itself at serine 1981 and then phosphorylates a number of downstream proteins (e.g. BRCA1, CHK2, H2AX, p53, NBS1 and SMC1) resulting in cell cycle arrest and the initiation of DNA damage repair. Loss of function of ATM is causal to a variety of syndromes involving increased incidence of several cancers.

Positive Control

Active Motif's HeLa nuclear extract (Catalog No. 36010) can be used as a positive control for ATM phospho Ser1981 antibody (Clone 10H11.E.12).
