Sirtuins are NAD-dependent deacetylases that are highly conserved among species. The original family member, Sir2 (silent mating-type information regulator 2) from yeast was identified as a gene involved in maintaining silent chromatin. In mammals, there are seven sirtuins (SIRT1-7) belonging to the class III histone deacetylase (HDAC) family of proteins. These are the only HDACs that require NAD for their enzymatic activity. Sirtuins play a role in metabolic diseases, neuronal physiology and aging, making them an important area of research.
