p300 antibody (mAb) tested by ChIP-Seq.
ChIP was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with chromatin from 4.5 million LNCaP cells and 5 µl of p300 antibody. ChIP DNA was sequenced on the Illumina HiSeq and 15 million sequence tags were mapped to identify p300 binding sites. p300 along with H3K4me1 are markers of active enhancer elements and are therefore expected to co-localize. A sampling of the p300 ChIP-Seq data shows the expected co-localization of p300 and H3K4me1.

ChIP was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with chromatin from 4.5 million LNCaP cells and 4 µg of p300 antibody. ChIP DNA was used in qPCR with the negative control primer pair or gene-specific primer pairs as indicated. Data are presented as Binding Events Detected per 1000 Cells using Active Motif's Epigenetic Services normalization scheme which accounts for primer efficiency and the amount of chromatin used in the ChIP reaction.

Background

p300 (E1A binding protein p300) functions as a histone acetyltransferase and regulates transcription via chromatin remodeling. Acetylates all four core histones in nucleosomes. Histone acetylation gives an epigenetic tag for transcriptional activation. Mediates cAMP-gene regulation by binding specifically to phosphorylated CREB protein. Also functions as an acetyltransferase for nonhistone targets. Acetylates 'Lys-131' of ALX1 and acts as its coactivator in the presence of CREBBP. Acetylates SIRT2 and is proposed to indirectly increase the transcriptional activity of TP53 through acetylation and subsequent attenuation of SIRT2 deacetylase function. Acetylates HDAC1 leading to its inactivation and modulation of transcription. Can also mediate transcriptional repression. Binds to and may be involved in the transforming capacity of the adenovirus E1A protein.
