The Active Motif High-Throughput ATAC-Seq Express Kit is designed to deliver rapid, reproducible, and scalable chromatin accessibility profiling in a true 96-reaction, high-throughput format. This kit integrates Pre-Indexed Assembled Tn5 Transposomes (also available separately as Cat. No. 53152) with the ATAC Buffer Set (also available separately as Cat. No. 53153), providing a fully optimized system for streamlined transposition and library generation. Pre-indexed transposomes eliminate the need for custom adapter loading and post-tagmentation indexing steps, enabling direct compatibility with multiplexed sequencing workflows and automated liquid handling platforms.
Q5 Polymerase and Library Purification (also available separately as Cat. No. 53159) is included to make High-Throughput ATAC-Seq Express a truly complete high-throughput ATAC-Seq solution for 96 cell or tissue samples including essential reagents for sample preparation, tagmentation, amplification, and library purification in a high-throughput-friendly protocol without sacrificing data quality.
The High-Throughput ATAC-Seq Express Kit is purpose-built for 96-well processing, utilizing the same principles as the ATAC-Seq Express Kit (Cat. No. 53157) and incorporating the power of the Pre-Indexed Assembled Tn5 Transposomes 96-well breakaway plate, eliminating separate pipetting steps for incorporating indexes, making it fully compatible with multichannel pipetting and robotic automation. The standardized format minimizes variability across samples, supports high sample throughput, and simplifies experimental setup—making it ideal for large-scale studies, screening applications, and core facility environments.

IGV Browser tracks of 50,000 cryopreserved K562 cells processed using the ATAC-Express Protocol (gold tracks) and the High-Throughput Protocol (purple tracks). Similar peak intensity and profiles between the different protocols confirm that the High-Throughput protocol is consistent with the ATAC-Express Seq Protocol. 50,000 fresh K562 cells were assayed in ATAC-Seq with either Active Motif’s Pre-indexed Assembled Tn5 Transposomes (top four tracks) or standard, non-indexed Tn5 Assembled Transposomes (bottom track). ATAC-Seq results were consistent between both methods. Browser tracks are shown at Chr6:159,668,373-159,821,466.

ATAC-Seq was performed on 2 replicates of 50,000 fresh K562 cells were assayed with Pre-indexed Assembled Tn5 Transposomes. i5 and i7 Indexed Primer combinations were optimized to yield consistent results. A subset of data is shown. The number of filtered peaks and FRIP score percentages were consistent across all index combinations.

100,000 cryopreserved HEK293, K562, HeLa, and GM12878 cells were assayed in ATAC-Seq with Pre-indexed Assembled Tn5 Transposomes resulting in consistent peak intensity as shown in this IGV browser view at Chr19:41,855,000-41,890,000.
