Background

SETD6 (SET Domain Containing 6, Protein Lysine Methyltransferase) is a member of SETD family, which has methyltransferase activity. This protein was reported to monomethylate ‘Lys-8’ of histone H2AZ, which is involved in nuclear receptor-dependent transcription. SETD6 was also reported to monomethylates ‘Lys-310’ of the RELA subunit of NF kappa-B complex, leading to down-regulate NF-kappa-B transcription factor activity. It may interact with several endogenous proteins which are involved in nuclear hormone receptor signaling. However, the real substrates remain unknown for a very long time.

Recombinant SETD6 protein is suitable for use in the study of enzyme kinetics, inhibitor screening, and selectivity profiling.

Protein Details

Full length human SETD6 protein (accession number NP_079136.2) was expressed in a baculovirus expression system and contains an N-terminal FLAG tag. The molecular weight of SETD6 is 52.1 kDa.

MTase-Glo Assay for Recombinant SETD6 protein methyltransferase activity
1 µM substrate peptide and 1 µM SAM were incubated with different concentrations of recombinant SETD6 protein in an 8 µl reaction system containing 50 mM Tris-HCl pH 8.6, 0.02% Triton X-100, 2 mM MgCl2, 1 mM TCEP for 1 hr. 5×MTase-Glo Reagent was added to the products and incubated for 30 min. Then MTase-Glo Detection was added and luminescence was read after another 30 min incubation. SAH standard curve (0-1 µM) was performed following the same protocol

Storage
