Background
SSRP1 (Structure Specific Recognition Protein 1), also known as FACT p80 (Facilitates Chromatin Transcription Complex 80 KDa Subunit), is a component of the FACT complex, a general chromatin factor that acts to reorganize nucleosomes. The FACT complex is involved in multiple processes that require DNA as a template such as mRNA elongation, DNA replication and DNA repair. During transcription elongation the FACT complex acts as a histone chaperone that both destabilizes and restores nucleosomal structure. It facilitates the passage of RNA polymerase II and transcription by promoting the dissociation of one histone H2A-H2B dimer from the nucleosome, then subsequently promotes the reestablishment of the nucleosome following the passage of RNA polymerase II. The FACT complex is probably also involved in phosphorylation of Ser-392 of p53/TP53 via its association with CK2 (casein kinase II).
Application Notes
This product was manufactured as described in Protein Details. Where possible, Active Motif has developed functional or activity assays for recombinant proteins. Additional characterization such as enzyme kinetic activity assays, inhibitor screening or other biological activity assays may not have been performed for every product. All available data for a given product is shown on the lot-specific Technical Data Sheet.
Protein Details
Recombinant human SSRP1 / FACT p80 protein was expressed in a baculovirus expression system as the full length protein (accession number NP_003137.1) with an N-terminal FLAG tag. The molecular weight of the protein is 82.2 kDa.
Storage
Recombinant proteins in solution are temperature sensitive and must be stored at -80°C to prevent degradation. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
