For optimal results, primary antibody incubations should be performed at 4°C. The addition of 0.05% Tween 20 to all Blotto solutions may also reduce background. Individual optimization may be required.

Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western Blot.

Immunogen

This NR2F2 / COUP2 antibody was raised against a peptide within the N-terminal region of human NR2F2.

Buffer

Purified IgG in PBS with 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

 

NR2F2 / COUP2 (pAb) tested by ChIP-Seq.
ChIP was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with chromatin from 4.5 million human endometrial stromal cells and 5 ul of antibody. ChIP DNA was sequenced on the Illumina HiSeq and 28 million sequence tags were mapped to identify NR2F2 binding sites. The purple track shows NR2F2 binding across a 1.2 million bp region on chromosome 12. Binding sites showed good correlation with the ENCODE active regulatory elements track (yellow).

Detection of NR2F2 / COUP2 by Western blot. The analysis was performed using 20 µg MCF-7 nuclear extract and NR2F2 / COUP2 (pAb) at a 1:500 dilution.

Background

NR2F2 / COUP2 (ARP1, TFCOUP2, COT2, COUP-TF2) is a member of the nuclear receptor superfamily related to the COUP transcription factor. It interacts with several nuclear receptor co-activator proteins to activate transcription. NR2F2 / COUP2 plays important roles in signaling pathways in multiple endocrine systems and is crucial for differentiation of the trophoblast during development.

Positive Control

Active Motif's MCF-7 nuclear extract (Catalog No. 36017) can be used as a positive control for NR2F2 / COUP2 antibody.

qPCR Primers

Active Motif's Human Negative Control Primer Set 1 (Cat. No. 71001) has been validated for performing qPCR and endpoint PCR when this antibody has been used for ChIP on human samples.
