Background

Nb2 nuclear extract was prepared from cell cultures of the rat lymphoma Nb2 nuclear cell line. The Nb2 cell line was derived from the lymphoma transplant in the thymus of a noble (Nb) strain male rat following prolonged estrogen treatment. Nb2 lymphoma cells are of pre-T cell origin and express the alpha and beta chains of the T cell receptor. However, they lack expression of class II MHC antigens. These cells are dependent on lactogens for mitogenic activity, which can also be stimulated by cytokines and human growth hormone and its analogs. Nb2 cells are most commonly used in studies of lactogen-induced mitogenesis and as a model for tumor progression of T cell cancers.

Extract Composition

Nb2 nuclear extract derived from rat lymphocytes was collected in Lysis Buffer. Lysis Buffer consists of 20 mM Hepes pH 7.9, 100 mM KCl, 20% glycerol, 1 mM MgCl2, 0.5 mM DTT and 0.5 mM PMSF. The protein content has been determined by a Bradford-based assay.

Storage

To ensure stability, extracts should be stored at -80°C.

We recommend aliquoting the extracts into single-use fractions and then storing them at -80°C. This eliminates repeated freeze/thaw cycles.
